Description DYNOtest anti-TPOn is a competitive radioimmunoassay for the quantitative determination of autoantibodies to thyroid peroxidase (TPO) in human serum using the coated tube technique. For the DYNOtest anti-TPOn BRAHMS employs exclusively enzymatic active native TPO from human thyroids. For the de- termination of anti-TPO antibodies older methods use as antigen denaturated resp. modified or recombinant TPO. In contrast to this the native structure of the antigen employed in the DYNO- test anti-TPOn guarantees the correct recognition of all classes and subclasses of antibodies, inclusive antibodies, which are directed only against the natural structure of the TPO. The anti- gen TPO used in this competitive assay is indirectly labelled with a monoclonal iodine-125 antibody to prevent the denaturation of the sensitive antigen. In connection with the monoclonal antibody on the solid phase this method guarantees a high sensitivity and specificity. During incubation, solid-phase monoclonal anti-TPO antibodies and enzymatic active native TPO from human thyroids form a complex with another iodine-125 labelled monoclonal anti-TPO antibody. Anti-TPO autoantibodies from a human serum sample or the standards compete with monoclonal anti-TPO antibodies immobilized on the inner surface of the tube for the binding sites on TPO. A high concentration of anti-TPO antibodies in the sam- ple leads to reduced binding of radioactivity to the inner surface of the tube. Therefore, the amount of radioactivity measured is reversely proportional to the antibody concentration of the tested sample. After the incubation period, the amount of human TPO and iodine-125 labelled anti-TPO antibody (tracer) not bound to the tube is removed by aspirating or decanting the liquid phase. The radioactivity remaining in the tube is measured. For anti-TPO antibodies there exists no international standard with official status.
