| Description | Human Insulin-like Growth Factor Binding Protein-1 (IGFBP-1) is a phosphoprotein, produced primarily in the liver, which specifically binds and modulates the bioactivity of IGF-I and IGF-II [1]. The role of IGFBP-1 has not been completely defined, but the nature of its regulation suggests that IGFBP-1 may be involved in glucose regulation and IGF-mediated substrate utilization [1,2]. IGFBP-1 has been shown to inhibit [3,4] as well as augment [5,6] IGF bioactivity. These paradoxical reports may be due to differential phosphorylation of IGFBP-1 which may significantly alter its affinity for IGFs and, therefore, its capacity to modulate their cellular responsiveness. As many as five IGFBP-1 variants, differing in their state of phosphorylation and relative expression in serum, amniotic fluid, decidua and cell culture media have been reported [7-11]. Non-phosphorylated IGFBP-1 has been shown to have 4- to 6-fold lower affinity for IGF-I compared with phosphorylated IGFBP-1 variants [7,9]. IGFBP-1 levels are inversely related to insulin concentrations and are not correlated with growth hormone or glucose levels [12,13]. Subsequently, measurement of IGFBP-1 levels may be useful as a marker of insulin action and hepatocyte function for research investigations.
