Description The test kit is an enzyme immunometric assay (EIA) in the microplate format with a liquid phase incubation, designed for the quantitative measurement of cortisol in serum. The microplate is coated with anti-rabbit antibodies. Calibrators and patient samples are added to anti-rabbit antibody-coated wells, along with rabbit anticortisol antibodies and ligand-labeled cortisol. During the 60-minute reaction, ligand-labeled cortisol competes with cortisol in the patient sample for antibody binding sites. During the same incubation, the resulting antigen/antibody complexes are bound to the well by the reaction with the immobilized anti-rabbit antibodies. The horseradish peroxidase-labeled anti-ligand is added to the wells. During a second 30-minute incubation, multivalent anti-ligand labeled with enzyme binds to the bound ligand-labeled cortisol. Nonreactive sample components are removed by a washing step. A chromogenic substrate, TMB (3,3',5,5'-Tetra-Methyl-Benzidine), is added to all wells. During a 30 minute incubation, the substrate is converted to a colored endproduct (blue) by the fixed enzyme. Enzyme reaction is stopped by dispensing of hydrochloric acid as stop solution (change from blue to yellow). The color intensitiy is inversly related to the concentration of cortisol present in the sample. The optical density of the color solution is measured with a microplate reader at 450 nm. Bi-chromatic measurement with a 600 - 690 nm reference filter is recommended.
