miRNeasy Mini Kit and miRNeasy 96 Kit

For purification of microRNA and total RNA from a wide range of animal tissues and cells

miRNeasy Kits allow purification of total RNA including miRNA or enrichment of miRNA in a separate fraction. The miRNeasy Mini Kit is suitable for low-throughput RNA purification. The miRNeasy 96 Kit allows high-throughput purification in a 96-well format.

Advantages of miRNeasy Kits:

  • Effective purification of total RNA including miRNA - from a wide range of animal tissues and cells

  • Efficient enrichment of miRNA - isolate RNAs <200 nucleotides

  • High-purity RNA - suitable for all downstream applications

  • Flexibility - flexible protocols for low-throughput or 96-well formats and for copurification or isolation of separate fractions

  • Flexible protocols for purification of total RNA or enrichment of miRNA
miRNeasy Kits enable purification of total RNA which includes RNA from 18 nucleotides (nt) upwards. The purified RNA is suitable for use in downstream applications such as northern blot analysis and quantitative, real-time RT-PCR. Alternatively, an miRNA-enriched fraction and a total RNA (>200 nt) fraction can be purified separately (see flowchart). A separate miRNA-enriched fraction may be required to reduce background in particularly sensitive downstream applications.

The miRNeasy Mini Kit is used for low-throughput RNA purification using spin columns. The miRNeasy 96 Kit enables high-throughput purification in a 96-well format, and is the first available 96-well format kit for miRNA purification.

For miRNA purification from formalin-fixed, paraffin-embedded (FFPE) tissue sections (using the RNeasy FFPE Kit), a supplementary protocol is available at .

Effective miRNA purification from animal tissues and cells
The miRNeasy Mini Kit and the miRNeasy 96 Kit efficiently purify RNA from animal tissues and cells, even when low amounts of starting material are used. In the results shown in Figure 1, CT values increased linearly when RNA was purified from decreasing amounts of cells or tissue, indicating effective miRNA purification from a wide range of amounts of starting material.

RNA can be purified from a variety of tissues and cells, including difficult-to-lyse tissues (Figure 2).

Purification of total RNA including miRNA allows direct comparison of miRNA expression levels with those of housekeeping reference genes or any other mRNA of interest (Figure 2).

    Efficient enrichment of miRNA
For some sensitive downstream applications, an miRNA-enriched fraction without larger RNA may be required. miRNeasy Kits enable efficient enrichment of RNA down to approximately 18 nt in size. In the data shown in Figure 3, the presence of very small RNAs was clearly visible after purification using the miRNeasy Kit, in contrast to samples prepared using an alternative kit.

    High-purity RNA
RNA prepared using miRNeasy Kits is highly pure and ready for use in sensitive downstream applications.

miRNeasy procedures eliminate the possibility of con tamination with salts or phenol which could interfere with later analyses (Figure 4). In addition to higher purity, miRNeasy Kits offer superior yields to alternative methods of miRNA purification such as using TRIzol Reagent (Figure 5).
