FULL SERVICE PROVIDER
Agencourt has a fully-automated genomic services pipeline based on SPRI, our patented magnetic bead-based nucleic acid purification technology. The high throughput nature of our pipeline allows for rapid 5 cDNA screening. Key cDNA clones identified during screening are easily integrated into our pipeline for high quality full length sequencing and finishing. All aspects of the pipeline are integrated and monitored by our Galaxy LIMS (Laboratory Information Management System) to ensure data quality.

SEQUENCING AND FINISHING
Purified cDNA clones are sequenced using BigDye Terminator v3.1 chemistry and reactions purified using CleanSEQ, a SPRI-based dye-terminator removal system. Typical Phred20 read lengths are greater than 550bp and pass rates greater than 85%. cDNA sequences are typically finished using primer walking. If a reference sequence is known, unique primer walking strategies allow for sample turnaround in less than two weeks.
The Mammalian Gene Collection (MGC) is an NIH-funded program aimed at identifying and sequencing cDNA clones containing a full-length open reading frame for all human and mouse genes. Agencourt was selected as the provider for all 5 EST sequencing of MGC cDNA libraries. Table 1 shows sequencing performance for over 15,000 EST reads conducted as part of the MGC initiative. Agencourt has submitted over 800,000 high quality cDNA sequences to GenBank as part of the MGC initiative.
