The QIAGEN LongRange PCR Kit contains all of the components required for the amplification of long PCR fragments. A unique blend of thermostable DNA polymerases, with enhanced processivity, allows the amplification of full-length PCR products. The enzyme mix provides high extension rates and proofreading ability for increased fidelity (up to 5-fold higher fidelity than Taq DNA polymerase). The novel buffer system provides highly specific long-range PCR, even from complex genomic DNA (Figure 1). The kit also contains nucleotides and Q-Solution an additive for the amplification of GC-rich templates. Preoptimized protocols make the QIAGEN LongRange PCR Kit easy and convenient to use.
A dedicated solution for long-range two-step RT-PCR
The QIAGEN LongRange 2Step RT-PCR Kit combines the features and benefits of the QIAGEN LongRange PCR Kit with a high-yield, full-length reverse transcription step. This complete and ready-to- use kit contains an enhanced viral reverse transcriptase, providing highly sensitive synthesis of full-length cDNA (up to 12.5 kb) even from low-abundance targets (Figure 2). The LongRange RT Buffer is specially formulated to provide efficient reverse transcription with no optimization required. The novel RNase H activity quencher binds to the cDNA:RNA hybridbinding site of the reverse transcriptase preventing degradation of the RNA during synthesis and enabling generation of full-length cDNA (Figure 3). The LongRange 2Step RT-PCR Kit also contains all of the components necesssary for subsequent long-range PCR.
Amplification of rare and difficult templates
The unique zwitterionic formulation of LongRange PCR Buffer improves pH buffering at high temperatures (7294C), minimizing pH-driven template degradation. In combination with Q-Solution (provided), it further facilitates amplification of GC-rich and other difficult templates.
