It took at least 100 pg of RNA to produce similar results using Supplier As nucleotides (lower panel). Each lot of PCR grade nucleotides is assayed for function in an RT-PCR amplification of a 1849 bp fragment of the human dystrophin gene. Varying amounts (5000, 1000, 500, 250, 100, 50, and 10 pg) of human skeletal muscle total RNA are incubated with 1 ml Titan RT-PCR System enzyme mix. After reverse transcription and 35 cycles of PCR, the reaction is analyzed on a 1.0% agarose gel. To pass the quality control test, a PCR product must be produced from < 100 pg of total RNA. Save money PCR-Grade Deoxynucleotides are supplied as sodium salts in highly purified water at pH 8.3 without the addition of additives or stabilizers. Since most amplification reactions are performed between pH 8 and pH 9, PCR-Grade Deoxynucleotides are compared to deoxynucleotides stored at pH 7 or 7,5 more resistant to chemical stress caused by pH shifts (Figure 3).
