Molecular Biology:Easy Dephosphorylation and Rapid DNA Ligation

Choose Shrimp Alkaline Phosphatase for Easier Heat Inactivation

Simplify dephosphorylation of 5′ ends on DNA vectors by using our high-quality Shrimp Alkaline Phosphatase. Avoid phenol:chloroform extractions; after dephosphorylation, inactivate Shrimp Alkaline Phosphatase completely and irreversibly by heating at 65 C for only 15 minutes.
Enjoy the convenience of a single-tube reaction; perform the restriction digestion, dephosphorylation, alkaline phosphatase inactivation, and ligation all in the same tube.

Ligate DNA in Five Minutes with the Rapid DNA Ligation Kit

Experience DNA ligation that is fast, flexible, and easy to perform with the original kit for rapid ligations.

  • Perform highly efficient ligations in 5 minutes at room temperature.
  • Ligate sticky- or blunt-end DNA fragments.
  • Generate constructs, transform competent cells, and plate bacteria all on the same day, letting you pick transformed colonies the next day.
  • Save time with a complete kit that contains enzymes and ready-to-use buffers.

Typical results: cloning of an insert into plasmid vectors

50 ng of pUC19 DNA, digested with Sma I or Hind III, respectively, and dephosphorylated with alkaline phosphatase, was ligated with 150 ng blunt- or sticky-end insert DNA, respectively, according to the standard protocol. The inserts were purified by electrophoresis, then isolated from the agarose gel by agarase digestion.
