( ...)GeneChip Sample Cleanup Module for cleanup of cDNA and in vitro ,,, transcribed cRNA

A crucial step in gene expression analysis is target labeling. To improve the critical steps in the target-labeling process, QIAGEN has developed the GeneChip Sample Cleanup Module specifically for use with Affymetrix GeneChip expression arrays. The GeneChip Sample Cleanup Module was developed as part of an ongoing collaboration between QIAGEN and Affymetrix, the market leader in microarray gene expression analysis. The GeneChip Sample Cleanup Module helps streamline and standardize preparation of target nucleic acids, to save time and effort in the lab and improve reproducibility (Figure 1).
Figure 1 Cleanup using the GeneChip Sample Cleanup Module was compared with a phenol/chloroform method previously used in GeneChip analyses. Brain, liver and HeLa total RNA (5 g) and yeast poly-A RNA (0.2 g) were used. A TaqMan -actin Control Reagents were used in real-time RT-PCR to quantify the amount of cDNA obtained before and after purification. Measurements were made in triplicate, and the mean value was used to calculate recovery. B After cleanup, 10 l of the 12 l of recovered cDNA was used for in vitro transcription. The cRNA yields were determined by UV spectrophotometry after adjustment by subtracting carryover RNA. Advantages of the GeneChip Sample Cleanup Module:
