Premium Quality siRNA Synthesis
Ambion's custom siRNA service provides the premium quality siRNAs you need for your research. Each siRNA is synthesized in our state-of-the-art chemical synthesis facility to meet the highest quality standards. Our standard purification procedure includes deprotection and column purification, and yields siRNA that is typically 90% pure (guaranteed >80% pure). For the most demanding applications, HPLC and PAGE purification is available, yielding siRNA that is guaranteed >97% pure.

To ensure that we are providing the highest quality siRNA, all HPLC and PAGE purified RNA oligonucleotides are analyzed for purity by capillary electrophoresis (CE) or HPLC. Furthermore, MALDI-TOF (matrix-assisted laser desorption ionization time-of-flight) mass spectrometry is used to analyze the molecular weight of each oligonucleotide (Figure 1). Finally, each annealed siRNA is analyzed by gel or capillary electrophoresis to ensure that the strands annealed properly.

Save Time with Highly Effective, Cenix Designed siRNAs
High quality siRNA is just one aspect of a successful gene silencing experiment. The design of the siRNA sequence is also critical. Ambion has partnered with Cenix BioSciences, which has developed a proprietary siRNA design algorithm that has been refined over the past year to yield an extremely high percentage of effective siRNA sequences (see Designing a Better siRNA). Figure 2 in that article demonstrates the high success rate of the algorithm. In a test of 79 siRNAs designed using the algorithm, 94% (74 of 79) reduced target mRNA levels by at least 70% when assayed 48 hours after transfection. You can now order siRNAs to your gene of interest that have been pre-designed using this algorithm. Multiple designs are immediately available for >95% of all human, mouse, and rat genes listed in the public RefSeq database maintained by NCBI. Ambion and Cenix are committed to continual refinements of the algorithm and corresponding updates to the pre-designed sequences as more is learned about siRNA design and as the RefSeq database is updated.


Complete Flexibility -- Choose One, Two or Three Pre-designed siRNAs Per Gene
To offer you the greatest flexibility in your research, you can have pre-designed siRNAs made at various synthesis scales, with your choice of purification method. Although a single pre-designed siRNA sequence has a high probability of reducing target mRNA levels, independently using two or more siRNAs to the same gene can help confirm the specificity of the siRNA effect. For that reason, Cenix created three designs for >95% of the human, mouse and rat genes in RefSeq. Order one, two, or three of these pre-designed siRNAs per target gene -- the choice is up to you. We can also use the algorithm to design siRNAs to genes in other organisms listed in the NCBI database, or to design siRNAs to target a sequence that you provide. And sequence information is included as part of the product information at no additional cost. Of course, you can also provide us with the siRNA sequence you want synthesized.
