The Calcium Plus Assay Kit provides a reliable and homogeneous method for detecting changes in intracellular calcium concentration. Specifically, this fast and simple fluorescencebased assay allows the researcher to accurately and easily detect intracellular calcium fluxes in targets that generate unreliable data using some traditional methods. Further, the Calcium Plus Assay Kit is designed for use with chemokine and small peptide targets, which can be problematic and produce less than optimal results with standard assay formats.


IMPROVED RESULTS
The primary advantage of the Calcium Plus Assay Kit is the improvement in the data quality. When compared with the standard fluo-3 AM dye incubation and wash procedure, results using the Calcium Plus Assay Kit are more consistent. Advances in the reagent chemistry and control of addition volumes are the pivotal reasons for the enhanced performance. These developments result in fewer spontaneous calcium fluxes and unresponsive cells. Figure 1 illustrates dose:response traces generated using the FLIPR384 system. Note the signal dynamic range and the small addition volume (6 L).

Strong signals are attained in cases that may previously have yielded no peak at all. When compared against assays using fluo-3, the peaks generated with the Calcium Plus Assay Kit are substantially higher and more distinct. Additionally, tighter CVs across 96- and 384- well plates are observed. Figure 2 illustrates the difference in kinetics and dynamic range between the equivalent concentrations of RANTES and MIP1a.


RAPID ASSAY DEVELOPMENT
With no wash steps, many of the artifacts of incubate and wash protocols are avoided. Cell lifting, lowered cell competency and incomplete dye washing are common problems that are easily avoided by using the Calcium Plus Assay Kit. The streamlined, homogeneous format also makes it less labor intensive to run. With the Mix-and-Read procedure, the cells can be incubated with the reagents and are stable for up to several hours. Rapid analysis of the cells can be followed with detection on a FLIPR system or FLEXstationmicroplate reader.


INCREASED THROUGHPUT
The Calcium Plus Assay Kit increases throughput by eliminating cumbersome wash steps. Less preparation time is required for wash buffers and wash calibrations. This successfully eliminates causes of variability in the data, resulting in fewer false positives and negatives. The kit is designed to work at room temperature, which is conducive to automation using stackers or robots.
