77.6.6 Sample cooling and subsequent PCR

If sensitive samples should remain cooled until the PCR takes place, the thermoblock must be preheated. This
can be carried out in two ways:

  1. Set the "Incu" menu to 4 C and "Heater is On".
  2. Adapt an initial program and connect via a "LINK" command.
    For this purpose, the temperature of the thermoblock is brought to 4 C using the first program. After the
    samples have been inserted into the cold thermoblock, the second program with the PCR protocol is
    called up via a LINK command.

Programming should take place as follows:

Program 1 e.g. COOL CNTRL TUBE
Lid
NOWAIT FIX
The entries in the first program are also valid for the programs which are added via LINK. The lid setting "Fix" must be selected. 1 Hold 4 C Enter

2. Link PCRPROT
end
The thermoblock cools until the processing program (e.g. PCRPROT) is started
via the LINK command by pressing .
Link to the second program with the PCR protocol. Program 2 PCRPROT CNTRL TUBE If the program also contains the "Tube control" command, further entries for
tube type and filling volume are no longer necessary; the entries made for the
first program are valid for all further programs.
