Description	The pWEB::TNC™ Deletion Cosmid Cloning Kit enables you to prepare, in about two days, a complete and unbiased primary cosmid library of genomic DNA for subsequent use in making nested deletion sublibraries. High efficiency cosmid library construction is accomplished using easy and highly reproducible kit protocols. First, genomic DNA is randomly sheared by passing it through a syringe needle. Next, the sheared DNA is end-repaired using reagents in the kit, and then size selected on a gel by comparison with a supplied 40-kb standard. Finally, the size-selected DNA is ligated into the supplied linearized and dephosphorylated pWEB::TNC Cosmid Vector, packaged using ultra-high efficiency MaxPlax™ Lambda Packaging Extracts (>10⁹ pfu/mg for phage lambda), also in the kit, and plated on the supplied EPBOS™ plating cells.
