| Description | The XpressPack Luminescent Detection System is intended for use in the quantitative detection of amplified DNA generated by an in-house validated method for in vitro nucleic acid amplification of a selected target of interest. • The XpressPack Luminescent Detection System is a hybridization capture assay designed to detect amplified nucleic acid specific to the target of interest. Amplification reactions use a biotinylated primer set (included in the XpressPackTM gene-specific kit) to generate the required amplified product (amplicon). A portion of the resultant amplicon is added directly to the capture plate (included in the XpressPack gene-specific kit) and denatured under alkali conditions to separate the two DNA strands. The single-stranded amplicon is captured to the plate via hybridization with a specific oligonucleotide probe immobilized to the reaction well (target specific probe-coated plate included in the XpressPack gene-specific kit). The hybrid complex is then reacted with streptavidin conjugated to AquaLite, a recombinant form of the photoprotein aequorin. Unbound conjugate is removed by washing. When calcium cations are added, a blue light is emitted. This light is quantified using a microplate luminometer; the intensity of the light is directly proportional to the amount of amplicon present in the sample.
