Description PolarScreen Glucocorticoid Receptor (GR) Competitor Assay Kits are ideal for studying or screening novel GR binding compounds. The PolarScreen GR Competitor Assay Kits contain partially purified GR protein, stabilizing peptide, buffer, and one of the following proprietary fluorescent glucocorticoid Fluormone ligands: GS1 (green assay kit), GS Red (red assay kit), or GS Far Red (far red assay kit). Fluormone GS Red and GS Far Red reduce interference from the intrinsic fluorescence found in some compounds when excited in the green region of the visible spectrum. This kit is designed for room temperature assays, using a coactivator-related stabilizing peptide to maintain GR complex integrity. The assay can also be run in the absence of the stabilizing peptide at +4C.To assay:protein added to Fluormone GS1, GS Red or GS Far Red forms a complex with a higher polarization value. Test compounds that compete for binding to GR and prevent the formation of a Fluormone/GR complex will result in a lower polarization value (Figure 1 and Table 1). The shift in polarization in the presence of test compounds is used to determine the relative affinity of test compounds for GR.

The PolarScreen Glucocorticoid Receptor Competitor Assay Kits contain partially purified GR protein, Fluormone GS1 (green assay kit), GS Red (red assay kit), or GS Far Red (far red assay kit) proprietary fluorescent glucocorticoid ligands, stabilizing peptide, and buffer. Store components as indicated in the assay protocol (-80C, -20, or +4C).
