| Description | Source: Crotalus adamanteus venom Description: Phosphodiesterase I (venom exonuclease) is widely used in studying nucleic acid structure and sequence(1). It hydrolyzes 5'-mononucleotides from 3'-hydroxy-terminated ribo-oligonucleotides and deoxyribo-oligonucleotides(2). Phosphodiesterase I cleaves ADP-ribosylated proteins at the pyrophosphate linkages to yield phosphoribosyl-AMP(3). Properties: Molecular Weight: 115,000 Da Optimum pH: 9.8 - 10.4 Activators: Mg2+ Inhibitors: 5mM EDTA, reducing agents (gluthathione, cysteine and ascorbic acid), partial inhibition by ATP, ADP and AMP Form: Lyophilized powder Purity: Partially purified by the method of Bjrk(4) and treated to remove contaminating 5' nucleotidase(5). Shipping and Storage: Shipped on dry ice. Store at -20C. Unit Definition: One unit hydrolyzes 1M p-nitrophenyl thymidine-5-phosphate per min at 25C, pH 8.9. Concentration: greater than or equal to 20 units/mg Applications: *Base composition and nearest neighbor analysis(1). *Removal of cyclic AMP from protein preparations. References: *HO, N. W.
