Description PROTECT FROM MOISTURE. AVOID FREEZE/THAW CYCLES. This kit allows the quantitation of apoptotic cells in vitro by DNA affinity-mediated capture of free nucleosomes followed by their anti-histone-facilitated detection. In this assay, mono- and oligonucleosomes are captured on pre-coated DNA binding proteins. Anti-Histone 3 (H3) biotin-labeled antibody then binds to the histone component of captured nucleosomes and is detected following incubation with streptavidin-HRP conjugate. HRP catalyzes the conversion of colorless tetramethylbenzidine to blue. The addition of stop solution changes the color to yellow, the intensity of which is proportional to the number of nucleosomes in the sample. By comparing the absorbance of unknown with that obtained from the standards, one can assign a nucleosome unit value to the unknown sample. Can be used to assign apoptotic index. One unit is defined as the amount of nucleosomes from 444 UV-treated Daudi cells.

Kit Components: 96-Well microtiter plate (12 removable strips) coated with a DNA binding protein, nucleosome standard, lysis buffer/sample diluent, biotinylated anti-histone 3 antibody, 400X HRP-streptavidin conjugate, conjugate diluent, chromogenic substrate, 20X concentrated wash solution, stop solution, 4 plate sealers, and a directional insert.
