| Description | Measurement of light production by luciferase-cDNA transfected cell lines has proven to be a valuable technique in the development of reporter gene assays in which the luciferase is used as an indicator for the transcription of other genes. The choice of the luciferase enzyme is due to advantages such as high sensitivity and relatively simple measurement. With traditional assay systems , signal measurement must be completed within a limited time period due to the short half-life of the reaction (half-life approximately five minutes). This means only one plate at a time can be prepared and counted. The PerkinElmer luclite Constant Quanta assay system produces a long lived glow type signal with a half-life of several hours, which makes it ideal for use on the PerkinElmer LumiCount Microplate Luminometer or the PerkinElmer TopCount Microplate Scintillation and Luminescence Counter. When performing high-throughput assays using luclite and the TopCount, you can prepare thousands of samples in microplates, load them into the microplate stacker, and analyze all samples unattended. The luclite assay system also provides the most simple procedure available - just mix two vials of reagent, dispense into the wells of a microplate containing transfected cells, seal the plate, shake briefly and measure. The signal obtained with luclite has a linear decay characteristic after an initiation phase of approximately one minute. Luclites long-lived luminescent signal produced is cell type dependent, but is generally stable for up to five hours. There are no additional cell lysis steps necessary when using luclite.
