Description The electrophoretic mobility shift assay (EMSA) is a powerful tool for evaluating DNA-protein interactions. Often referred to as gel shift or gel retardation, the assay is based on the principle that when subjected to electrophoresis, free DNA will migrate differently than a DNA-protein complex. The Pierce LightShift Chemiluminescent EMSA Kit is a nonradioactive system for performing EMSA using Pierce's proprietary chemiluminescent detection technology. Biotin-labeled DNA can be prepared using the Pierce Biotin 3’ End DNA Labeling Kit (Product# 89818) or ordered directly from your custom oligo supplier. Capable of sub-femtomole detection levels, the LightShift Chemiluminscent EMSA Kit offers the sensitivity and speed of radioactive assays without the hazards, waste and probe instability problems associated with radioactive systems. It comes with all necessary components, except positively charged nylon membrane, for performing 100 DNA-protein binding reactions and sufficient chemiluminescent detection reagents for 800 cm2 of membrane. A detailed protocol for optimizing EMSAs as well as a troubleshooting guide are also provided.
