Description QED Bioscience has over 23 years of experience in the development of hybridoma cells lines. We have made antibodies to large and small peptides, proteins, haptens, carbohydrates, bacterial and viral antigens, and plasmid DNA-encoded antigens. We understand and appreciate that every antigen is unique, so our hybridoma development programs are never "cookbook". We will tailor a program to your needs after we discuss important features of your antigens and the antibodies you require.

Features and Benefits:

  • Typical time frame for completion is 5-6 months.
  • Project tailored to the clients specific antibody needs.
  • Our programs are "modular", so you can choose how large or small you want each phase of your program to be.
  • Supernatant samples available to the client for testing in their system for subcloning selection.
  • Antibody is produced from 5 clones selected by the client; antibody from additional clones can be produced for a nominal cost.
  • All materials are the property of the client and are maintained until directed otherwise by the client.
  • Development is set up in phases. Billing is at the beginning of each phase, so if the project does not proceed to the subsequent phase, you are not billed.
PHASE I: IMMUNIZATION

1. Immunization of 5 Balb/c mice.
2. Initial rest period of 8 weeks to optimize for high titer IgG responses.
2. Test bleeds are evaluated by ELISA; bleeds are available to the client for testing.
3. The animals with the highest titers for selected for fusion.

Estimated completion time: 10-14 weeks ($1,845.00)

PHASE II: FUSION

1. Spleen cells from selected animals are fused with two different myeloma cell lines.
2. Remaining fusion cocktail is frozen for future use, if necessary.

Estimated completion time: 2-3 weeks ($3,690.00)

PHASE III: ANALYSIS

1. Antibody-positive parental hybridomas and subclones are evaluated by ELISA.
2. Supernatant fluid samples are available to the client for evaluation in their systems.
Up to 25 antibody-positive cultures will be carried through the subcloning and analysis phase.
5 clones may be selected for Antibody Production; antibody may be produced from additional clones for a nominal charge.

Estimated completion time: 6-8 weeks ($2,240.00)

PHASE IV: ANTIBODY PRODUCTION

1. Antibody is produced either in ascites (5 mice per clone) or in vitro (150 ml per clone).
2. Our proprietary in vitro medium produces 5-25 fold greater yields of antibody than conventional media.
3. All antibody production is done to cGMP standards.

GMP Ascites Production

QED offers complete programs for ascites production, from small R&D scale to pilot lot production to large-scale production. We are a USDA-registered research facility, and we comply with all federal guidelines for care and use of laboratory animals.

You may provide your hybridoma cell lines to QED either frozen or in culture. We will advise you on how many cells to send and how to ship your cells to us. Approximately one week is needed to prepare cells from frozen stocks for injection, and there is an additional charge for this service (see below).

Nude and SCID mice are available for ascites production for non-murine hybridoma cell lines.

Hybridomas are injected into pristane-primed mice, then mice are monitored daily for ascites development and general condition. Ascites collection begins when accumulation of ascites becomes evident and continues until final harvests are performed.
