Description The HEEBO (Human Exonic Evidence Based Oligonucleotide) Genome Set and MEEBO (Mouse Exonic Evidence Based Oligonucleotide ) Genome Set contain a collection of open source oligo probes, largely derived from constitutively expressed exons, allowing interrogation of thousands of genes. Developed in collaboration with leading researchers at Stanford University and UCSF, the sets enable the study of human and mouse transcription patterns, and as broadly as possible, alternative splicing. An exon-centric design allows the differentiation of constitutively expressed versus alternatively expressed exons. This design also supports comparative genome hybridization (CGH) analysis. In addition to the exon-centric probes, the set contains an extensive assortment of controls that facilitate accurate evaluation of expression results. Probes are Tm-normalized and are designed to minimize cross-hybridization and secondary structure.

  • Contents and Storage: Each HEEBO Genome Set contains 48,958 70mer 5 amino modified oligonucleotide probes in 128 384-well plates. The set includes 44,308 probes targeting human genes, 864 positive control probes, 410 negative control probes, and 2,733 doped control probes. Each MEEBO set contains 38,467 70mer 5 amino modified oligonucleotide probes in 101 384-well plates. The set includes 35,302 probes targeting mouse genes, 1,152 positive control probes, 97 negative control probes, and 1,916 doped control probes. Probes are provided at a final yield of 200 or 600 pmol each and delivered in microarray print plates (Genetix model X7020). Each order also receives a data-CD with sequence information. For maximum stability, dried oligos should be stored at -20C with minimal exposure to light. Plates should be handled carefully to minimize the possibility of bacterial contamination. Repetitive freeze/thaw cycles could result in probe degradation. Special care should be used when removing plate seals to resuspend the oligos; plates should be spun down prior to removing seals, and seals should be removed slowly.
