Description The Hamamatsu C9136 FLIM system combines the space and time dimensions of a streak camera image with the resolution of the optical microscope to open new horizons in biological imaging. Using a short, pulsed light source for excitation, the decay of a fluorescence emission can be captured and the lifetime of the decay measured. Subtle changes in the local environment of a fluorescent molecule such as pH, ion concentration, polarity and viscosity produce changes in the lifetime of the fluorescence decay. Molecular non-radiative energy transfers (FRET) also cause detectable changes in the lifetime. Unlike intensity-based measurements of fluorescence, the lifetime (tF) is constant even if concentration, path length or illumination is varied in the sample. Minimal excitation levels, very fast sampling, and tremendous sensitivity to changes in lifetime make this and ideal system for biological investigations in living cells.
