Description Useful as a positive control for ChIP assays. Efficient fixation of a protein to chromatin in vivo is a crucial step for the Chromatin-immunoprecipitation (ChIP) technique. The extent of crosslinking is probably the most important parameter. Two major problems concerning the subsequent immunoprecipitation step should be taken into account: first, an excess of crosslinking can result in the loss of material or reduced antigen availability in chromatin, or both, and second, the relative sensitivity of the antigen epitopes to formaldehyde. For rapid checking chromatin IP efficiency. DNA from an aliquot of IP-chromatin solution can be first reverse crosslinked and purified by phenol extraction. Then, PCR reactions using specific primers to amplify certain known promoter regions can be performed to check whether the chromatin immunoprecipitation is successful.
