| Description | Sac-Cel (Second Antibody Coated CELlulose) was developed in the late 1960s by Wellcome Diagnostics of Dartford, Kent, one of the earliest commercial pioneers of immunoassay in the world. IDS acquired the product range in 1987. Sac-Cel rapidly found favour with researchers and clinical biochemists developing their own "in house" radioimmunoassays, and became a benchmark methodology for the separation of Bound from Free fractions (B/F) in RIA. When used in liquid phase immunoassays, Sac-Cel provides a ready-to-use solid-phase separation medium, fine enough to remain in suspension during pipetting stages, but delivering firmer pellets in much shortened centrifugation time. After incubation with primary antibody (in the absence of potentially interfering carrier serum) and tracer, Sac-Cel is added. Following a second incubation, the reaction volume is increased by dilution prior to a short, low speed spin (typically 2 minutes at just 1,000 g). This effectively washes the solid-phase during separation, reducing background signals (NSB; non-specific binding). The resulting firm, very visible pellet facilitates aspiration or decantation, resulting in improved precision and the enhanced sensitivity that this confers. Sac-Cel has been formulated to address the IgG fraction of the major host species for primary antiserum development, namely rabbit, sheep/goat, guinea pig, and mouse/rat. Sac-Cel has also been used for affinity purification, and a host of other laboratory applications not formally reported to us! Sac-Cel has also been incorporated into many commercial RIA products by clients in the USA, Germany and Scandinavia. We also use it in our own Vitamin D assays. |
