Description ABsolute QRT-PCR Mixes are formulated for use in one-step QRT-PCR reactions, with gene-specific primers and SYBR Green. One-step QRT-PCR mixes offer a quick and simple method for analysing RNA. The pipetting steps are minimised, making the protocol easy to follow and automation possible. The 2x ABsolute QRT-PCR Mix is supplied with a patented blend of RTases combining the advantages of AMV and MMuLV RTases to give unparalleled performance. The mix can be used at elevated temperatures, avoiding problems caused by secondary structure in the RNA template. In addition the QRTase Blend has greater dynamic range than either AMV or MMuLV. This leads to greater accuracy in QRT-PCR experiments. Unique QRTase Enhancer - Provided to reduce problems associated with DNA contamination. ABsolute QRTase Blend - ABsolute QRT-PCR kits contain a unique blend of MMuLV and AMV RTases in a buffer designed for synergy with subsequent QPCR reactions. This allows detection of rare templates. The RNase H activity facilitates the freeing of priming sites during the QPCR stage of the reaction. The AMV component allows reverse transcription to be carried out at up to 57C when secondary structure is problematic. A RNase inhibitor is included in the blend to prevent RNA degradation from RNase A. Optimised Buffer - ABsolute QRT-PCR Mixes are based on a balanced buffer to maximise performance of both the QRTase Blend and Thermo-Start Polymerase. SYBR Green I is also included in the buffer at an optimum concentration to facilitate detection of low abundance templates. Maximum Specificity - Thermo-Start DNA Polymerase remains completely inactive during the reverse transcription stage of QRT-PCR. Enzyme is fully activated by a 15minute activation at 95C.
