Polytene


Polytene chromosomes are found in salivary glands of Diptera such as Drosophila. Their 2 mm length is much longer than metaphase chromosomes.
The polytene chromosomes consist of 100 or more copies of DNA, arranged side by side.

The term proposed by Koller (1935) to describe the multistranded condition (Gk. polys, many; tania, ribbon) of the chromosomes found in certain specialized tissues such as the salivary glands of Drosophila.

Polytene chromosomes are subdivided into some 5,000 dark bands separated by light interbands. Genes are located in both, but those in the interband regions seem to be more active.
The boundaries between the bands and interbands contain insulators.

The mature larvae show giant chromosomes in the salivary glands called polytene chromosomes - "puffs" indicate regions of transcription and hence gene activity.
It has only 4 pairs of chromosomes: 3 autosomal, and 1 sex.
