Multiple Cloning Site

Multiple Cloning Site: An artificially constructed region within a vector molecule which contains a number of closely spaced recognition sequences for restriction endonucleases. This serves as a convenient site into which foreign DNA may be inserted.

Most also contain a polylinker or multiple cloning site (MCS), which is a short region containing several commonly used restriction sites allowing the easy insertion of DNA fragments at this location. See also 'Applications of plasmids', below.

A synthetic version of the phage f1 origin of replication is located a short distance upstream of the multiple cloning site.

Cloning vectors minimally contain an origin of replication, selectable marker gene (e.g., ampicillin resistance gene), and multiple cloning site containing unique restriction enzyme sites; other useful features may also be present.

Cloning site A restriction enzyme site or group of sites (= multiple cloning site) located in a vector at the best position for insertion of DNA.
Colony lift Blot taken from colonies (normally bacterial) growing on an agar plate.
