Origin of replication

Origin of replication
Discrete positions on a chromosome or vector DNA where replications are initiated. Vectors must have at least one origin of replication sequence for their replication in host cells.

Origin of replication (ori)
The nucleotide sequence where DNA replication is initiated. Determines the specific position on a DNA molecule where DNA replication begins.

origin of replication
A specific sequence of bases in a nucleic acid molecule to which the enzymes responsible for replicating the nucleic acid bind to initiate the copying process.

Origin of replication: A site at which DNA replication is initiated. There is only one in bacterial chromosomes, but numerous origins in eukaryotic chromosomal DNA. (Abbr. "ori") ...

In the initiation step, several key factors are recruited to an origin of replication. This origin of replication is unwound, and the partially unwound strands form a "replication bubble", with one replication fork on either end.

This works fine for the 3' -> 5' strand of a chromosome as the DNA polymerase can move uninterruptedly from an origin of replication until it meets another bubble of replication or the end of the chromosome.

Every plasmid contains at least one DNA sequence that serves as an origin of replication or ori (a starting point for DNA replication), which enables the plasmid DNA to be duplicated independently from the chromosomal DNA (Fig. 2) ...

Cloning vectors minimally contain an origin of replication, selectable marker gene (e.g., ampicillin resistance gene), and multiple cloning site containing unique restriction enzyme sites; other useful features may also be present.

Once the polymerases have opened the molecule, an area known as the replication bubble forms (always initiated at a certain set of nucleotides, the origin of replication).
